Kampot is on the Phnom Penh-Takeo-Sihanoukville railway line. Some trains run on this line over the weekend. More about it here

By bus

From morning to early afternoon buses run from Phnom Penh to Kampot (Sorya, Capitol, Hua Lin ... $ 4-5, 4-5 hours). Otherwise there is the possibility to take the bus to Sihanoukville and continue from there with a shared taxi.
